Esther Audu was born as Esther James Audu on March 22, 1982, in Ikeja, Lagos, to Mr. James Audu, a retired military officer who spent the majority of his career in Lagos and lived at the Ikeja Military Cantonment where Audu was born, and her five other siblings.

Audu is the youngest of six children from Olamaboro in Kogi State, and she attended primary and junior secondary school in Lagos. In 2002, their family relocated from Lagos to Abuja, where she completed her secondary school education before enrolling in Business Management at the University of Jos in Plateau State in 2006. She graduated with a BA in Business Management in 2010.

Esther began her acting career in high school and stated that her dream was to be a newscaster. She was a member of the Drama and Literary clubs in secondary school, where she performed in stage plays.
However, in 1996, she was chosen to represent Nigeria at Kidafest in Ghana, and her passion for acting began to grow, and she abandoned her dream of becoming a newscaster.

She first appeared in a lead role in Ungodly Romance and Sins of Rachael in Jos by Alex Mouth, which she described as the first films that helped launch her career fully into Nollywood. She appeared in a movie called Fatal Mistake as a sub lead while she was still an undergraduate.
